If there are any HD texture packs for the port, I am unaware of any (although, mods are being made). Otherwise, you start off with the vanilla N64 textures, which still look pretty decent by default at 1080p and up (UI on the other hand, you'll just have to put up with, tinker with the filtering settings to see what is most tolerable).
To start off with, you have mouse support, modern controller support, HD/4K resolution with widescreen, high framerate unlock (your mileage varies past 120 FPS), native splitscreen support, and a whole lot of customizability within the options menu (a new category not found on N64 "Extended") which includes an option to unlock everything out of the gate (mainly for multiplayer purposes).
There is also a mod out there that allows for online play (via simulated splitscreen). I'm blanking on the name, but you should be able to find it via the YouTube channel Graslu00, specifically his recent PD multiplayer livestreams.
